Methanol wird in einem Fixierungsprozess verwendet, der bei der Analyse
von Proteinen und DNA/RNA in Polyacrylamidgelen erforderlich ist. Diese Fixierung ist
notwendig, um Proteine oder Nukleinsäuren in große unlösbare Aggregate
umzuwandeln, die nicht reagieren oder zerfallen können. Ethanol ist ein
gutes Substitut für Methanol, da es weniger toxisch und die Arbeit mit
dem Stoff somit sicherer ist.

Die Substitution funktioniert bei der
Western Blot Analyse und der Aufbereitung von Membranen nicht gut. Es
sollte daher nicht für PVDF-Membranen verwendet werden.

Substitution of methanol with ethanol for use in fixation of polyacrylamide gels in analysis of proteins and DNA/RNA.

Methanol is used in a fixation process required for analysis of proteins and DNA/RNA on polyacrylamide gels. Fixation is necessary for transforming the proteins or nucleic acids into large, insoluble aggregates which are unable to react or degrade. Ethanol is a good substitute for methanol since it´s less toxic and therefore safer to work with.

The substitution does not work well with Western blot analysis's and processing of membranes. It should also not be used on PVDF-membranes.

Formamide is used to denature DNA in Fluorescent In Situ Hybridization (FISH). The specific purpose of the formamide is to make the DNA in the sample single stranded, which allows a probe to bind into the remaining strand. The FISH technique is a method used to locate the presence or absence of specific sequences of DNA in chromosomes.

For the past 15 years the department for clinical genetics at Sahlgrenska University Hospital has substituted the 70% solution of formamide with 30 ml of 1.9% solution of sodium hydroxide and 70 ml of absolute ethanol (99,5%). The substitution works well and the staff has not noticed any specific limitations with the alternative.

Certain pharmaceuticals including some cytostatics are carcinogenic, mutagenic, teratogenic and fetotoxic. Cytostatics and their metabolites are excreted by patients into waste water. In laboratory testing, the frequently used isomeric anti-tumour agent Ifosfamide was shown to be non- biodegradable. It is not eliminated in municipal sewage treatment plants, and therefore passes into the effluent. Structural modified compounds like Glufosfamide are still active against tumours, and in contrast to Ifosfamide are inherently biodegradable.
